I'm planning to construct a 16x12x9.5 foot greenhouse in my backyard, but there's an overhead power line coming from a nearby pole that runs directly into my home. I'm wondering if this completely rules out my plan to build under the line, or if there's any way I can still make it work. My yard is pretty much just an empty space filled with grass, and I want to do something productive with it!

You might be in the clear if the greenhouse can't be easily accessed from above. Generally, they require a good clearance—I'd recommend at least 3 feet. When I built mine, I called up the city for confirmation, and they told me it should be 200 square feet max, which is perfect for your size! Go for it!
As long as the greenhouse isn't easy to climb on top of, you should be fine. It's best to check in with your city for more specific guidelines. If you're still concerned, consider installing a pole on your house that would allow the line to be lifted higher. I did something similar at my last home and it worked out great—just make sure to discuss it with your utility company first! Also, if the line is already about 11-12 feet up, just keep your greenhouse closer to the house so it doesn’t intersect with the line's lowest point.
You should check the easement for the power line, which details any restrictions about building under it. This is super important to ensure everything's safe and compliant.
