I'm trying to figure out how to apply wipe-on polyurethane to a 2x4 on all four sides without any drilling or hanging options. Does anyone have tips on how to do this effectively?

You might want to try using painter's pyramids! They’ll allow you to elevate the 2x4 a bit and leave minimal marks once the finish is dry.
One method is to just take your time. Do one side, let it dry completely, and then flip it over to do the next side. It doesn’t rush things and you’ll get a nice finish.

Another option is using 3-inch screws on each end and resting them on saw horses. This way, you can do all four sides without holding it yourself.
