I'm planning to replace my two 30" open cavity vanities with a single 60" vanity, but I've noticed that the water lines and drains are misaligned. How challenging is it going to be to re-plumb the lines? I'm fairly handy and think I can handle it, but should I go all in on re-plumbing everything, or should I consider backtracking and just get two new 30" vanities instead?

You can't just have an open drain in the wall, so you'll need to address that. You can run a new drain line from the existing hookup to the new sink—just remember that it needs to slope down after the p-trap. For the water line, a long flex line might do the trick!
