I'm running a doorbell wire through my wood siding and need to drill a 1/4" hole for it. To properly seal and insulate the hole, should I use silicone caulk or low expansion spray foam on both sides of the hole? What would work best to trap the wire and ensure a good seal?

I'd recommend doing both! Use caulk first for an airtight seal and then fill it in with spray foam to ensure you get a good fit around the wire. That way you're sealed nicely on both sides.
For a hole that size, caulk should do the trick. It can fit nicely in a 1/4" hole and should provide a decent seal. Start with caulk on the exterior and you should be good!
