I recently installed a hose reel on my brick wall, but while drilling the 2" deep holes for the anchors, I think I accidentally drilled through the brick in two spots. I managed to secure the anchors and screws, and the hose reel seems stable. Should I worry about waterproofing these holes with caulk to prevent any moisture issues, or is the installation solid enough with just the anchors and screws?

You're actually okay! Brick walls usually have weep holes at the bottom to help with drainage, so as long as you’ve got good anchors, it should hold just fine without any extra waterproofing.
Yup, just anchor it securely! The screws and anchors should be enough support. Remember, brick is designed to let water escape, so you shouldn't have any issues down the line!
Caulking is a good idea, especially if you're concerned about moisture. I recommend using polyurethane caulking—it's stickier than silicone and really seals up well. Just make sure to clean the area first with some isopropyl alcohol. You can apply it around the hose reel mount if you want that extra layer of protection.
